817 Encapsulated activated-grape seed extract (E-AGSE): A novel liposome-based formulation that promotes anti-aging, brightening and hydration in human skin

Protein phosphatase 2A (PP2A) is a critical target to control cumulative oxidative stress and chronic inflammation during skin aging by reversible methylation of the C-terminal leucine PP2A catalytic subunit (PP2Ac). A novel, proprietary Activated-Grape Seed Extract (AGSE), enriched for PP2A-activating flavonoids, was recently developed, has demonstrated potent antioxidant anti-inflammatory activities. AGSE dark, purple-colored powder with limited solubility, which restricts its use in broad range formulations. To combat these restrictive properties, we developed an encapsulation formulation that both reduced color increased solubility AGSE, making it more amenable application wider array products. Encapsulation performed utilizing liposome hydroxypropyl-β-cyclodextrin, (HPCD)-based approach produce Encapsulated (E-AGSE). Human dermal fibroblasts epidermal keratinocytes were used determine expression levels dermal–epidermal junction (DEJ) markers. EpiDerm™ UVB-irradiated measure effects against cytokine release, DNA damage, apoptosis, barrier. We E-AGSE inhibits demethylation, increases key anti-aging (collagen I, III, elastin) DEJ markers, protects UVB-induced damage reduces inflammation.
